1. Platelet-vessel wall interactions
Specific aims:
To understand the interaction of vWF with collagen at the atomic level.
To understand the conformational changes that convert vWF into its GPIb binding conformation.

2. Auto-immune induced thrombosis
Specific aims
To understand why antibodies directed against 2Glycoprotein can cause thrombosis in every vessel within the human body.
To understand the role of LRP8 plays in haemostasis.
To understand the consequences that conformational changes within vWF and GPIb have for the development of thrombotic microangiopathies (but sometimes also haemorrhages).

3. Interaction between haemostasis and inflammation
Specific aims:
To understand the role of 2-glycoprotein I, vWF and platelets in inflammation and innate immunity

4. Interaction between clotting factors and platelets
Specific aim
To understand why factor VIIa prevents bleeding in patients with a platelet function defect.

5. Diagnostic assays to detect an increased risk for thrombosis
Specific aims:
To improve the diagnosis of the antiphospholipid syndrome.
To improve the diagnosis of thrombotic angiopathies with an assay that recognises vWF in its GPIb binding conformation.
To develop an assay that detects levels of factor VIIa
To develop biomarkers for progression of atherothrombotic disease based on the information that is present in circulating platelets.
